More precisely, the invention is directed towards a method that decreases the pH value of Cremophor-EL using clay or metal salts and provides improved Cremophor-EL. The invention also provides formulations using such improved Cremophor-EL. |
Full Text | WE CLAIM: 1. A method of purifying polyethoxylated castor oil, comprising the steps of: (a) contacting untreated castor oil with a pH reducing agent in the presence of a solvent at temperature ranging between 20 to 40°C for a period of about 5 to 12 hours, and (b) obtaining purified castor oil having decreased pH and low ionic content. 2. A method as claimed in claim 1 wherein the untreated castor oil has a pH of 6 to 8 with viscosity in the range of 600-800 units. 3. A method as claimed in claim 1 wherein the pH reducing agent is selected from clay, metal salts or a combination thereof. 4. A method as claimed in claim 3 wherein the metal salts are alkaline or alkaline earth metal salts, such as potassium hydrogen sulphate, copper sulphate, iron sulphate. 5. A method as claimed in claim 3 and 4 wherein the ratio of metal salt to clay is preferably 1:1 to 1:0.5 with respect to polyethoxylated castor oil. 6. A method as claimed in claim 4 wherein the concentration of clay or metal salts is 1 to 10% w/w with respect to polyethoxylated castor oil. 7. A method as claimed in claim 1 wherein the untreated castor oil is mixed with a pH reducing agent in the presence of a solvent. 8. A method as claimed in claim 1 wherein the solvent is selected from ethanol, isopropanol or butanol, preferably ethanol. 9. A method as claimed in claim 1 wherein the preferred ratio of castor oil to solvent is 1:1 or 1:2 (wt/vol). 10. A method as claimed in claim 1 wherein castor oil is preferably purified and solvent removed under vacuum. 11. A method as claimed in claim 1 wherein purified castor oil has a viscosity decrease by 100 to 150 units. 12. A method as claimed in claiml wherein the purified castor oil has a pH of 3.5 to 4.5. 13. Stabilized and purified castor oil obtained by the method of claims 1 to 12. 14. A composition useful for treatment of cancer comprising an anti-cancer agent and a pharmaceutically acceptable amount of polyethoxylated castor oil purified by the method of claims 1 to 12. 15. A composition as claimed in claim 14, wherein the anti-cancer agent is paclitaxel. 16. A method of purifying polyethoxylated castor oil substantially as herein described with reference to the examples. 17. Stabilized and purified castor oil substantially as herein described with reference to the examples. 18. A composition useful for treatment of cancer substantially as herein described with reference to the examples. |
