| Abstract | The present invention relates to; A process for the synthesis of a synlt1etlc polypeptide analog of parathyroid ~e (PTH) or parathyroid homme related peptide (PTHrP). or a salt there-of, In which amino acid residues (22-31)selected from (SEa 10 NOS: 85. 86,26,27,28,29, and 30) form an amphtpathica-hellx, which processoompr1ses: a) independently synthesizing three precU1'8Or peptide fragments: an N-termlnus, a mkjdle. and a C-terminu6 fragment, wherein the N-terminus fragment has a C-termlnal glycine, the middle fragment has a C-terminal leucine, and the C-terminus fragment has an N-tem1lnalleuclne. b) cleaving all but the I1tended ultimate c..termlnal precursor peptide fragment of the des/red polypeptide from their respective resn suppOrts; c) sequentially condensing said cleaved precursor peptide fragments with the resin bound c..terminal peptide fragment to form the desired polypeptide product; d) removing side chain protecting groups; and e) cleaving the polypeptkje prodoot from the resin support. |
