The instant method can be used for in-process control in the production of rabies vaccines. The instant method is highly reproducible, sensitive, specific and rapid as compared to conventional ELISA & SRID methods. |

The following specification particularly describes the nature of this invention: 14 AUG 2008 24.00 ESTIMATION OF RABIES GLYCOPROTEIN The ' quantitation of rabies glycoprotein is required for various samples containing varying, high or low, concentration of the glycoprotein. The single radio-immunodiffusion (SRID) test is not a very sensitive method to detect antigens at low concentrations. The conventional ELISA has got a higher sensitivity than SRID but is not sufficient for the lowest levels of glycoprotein obtained in regular harvests of cell culture. Both the methods are time consuming. There are published methods available for estimation of antibodies but highly sensitive, specific and rapid method for estimation of glycoprotein is not found in literature or practice. The instant invention relates to a method of estimation of rabies glycoprotein which is highly sensitive, specific and rapid. Summary of the invention: The polystyrene carboxylated microspheres (beads) are coupled with anti-rabies monoclonal antibody. The beads are subjected to reaction with rabies glycoprotein samples. After washing, the beads are treated with anti-rabies antiserum raised in species other than mouse e.g. sheep. After washing, the beads are treated with fluorescent conjugate e.g. anti-sheep IgG conjugated to Phycoerythrin. After washing, the beads are analyzed by Bioplex 2 00 or similar system and the concentration of rabies glycoprotein is estimated. The method detects antigen even at 3.125 mlU/ml concentration and the incubation time is about 2.5 hours only. |
